I would suggest a compression check and a leak down check. After that measure the valve lift on the affected cylinders against what cam you have. This can be done with an indicator. Mine turned out to be a flat lob and grooved lifter on number 5 exhaust valve. Had to replace the cam, lifters, valve seals, push rods, and rocker arms. Though mine was just the misfire you quite possibly have a blown head gasket. Do the compression and leak down first.
